How a skull came to mean laughter
The literal meaning is death, danger or poison, and the emoji was added in Unicode 6.0 in 2010 with that sense. Its dominant use today is entirely different: it stands in for 'I'm dead', internet shorthand for laughing so hard you have expired.
That shift happened largely on Twitter and TikTok in the late 2010s and arrived on Discord with the same users. It has now displaced the laughing emoji as the default reaction to something funny among younger users, to the point where the older emoji reads as earnest by comparison.
The literal reading has not disappeared - in a gaming context a skull still marks a death or a defeat - so it is one of the few emoji whose meaning genuinely depends on who is reading it.
Not to be confused with the crossbones
There is a separate Skull and Crossbones emoji at U+2620, which retains the unambiguous danger and poison meaning and never picked up the humorous sense. The two are visually similar at small sizes and are frequently swapped by mistake.
If you mean danger, the crossbones is far less likely to be misread. If you mean laughter, the plain skull is the one everyone recognises.
In server configuration
Servers with a younger membership frequently use it as their starboard emoji instead of the star, because it is what members already react with. That makes the highlights channel self-populating rather than requiring a deliberate act.
In gaming servers it marks death-count channels, hardcore-mode roles and permadeath trackers, where the literal meaning is the intended one.

The crossbones version is a different character entirely rather than a variant, which is why searching for skull in the picker returns both and it is easy to pick the wrong one.

The humorous reading is entirely a product of late-2010s social media and attached only to the plain skull, leaving the crossbones with its original sense intact.
